क्लोपेरास्टाइन (INN) या क्लोपेरास्टिन, क्लोपेरास्टाइन हाइड्रोक्लोराइड (JAN) (ब्रांड नाम हस्टाज़ोल, निटोसिल, सेकी) और क्लोपेरास्टाइन फेंडिज़ोएट के रूपों में, एक एंटीट्यूसिव (खांसी रोकने वाली) और एंटीहिस्टामाइन है जिसे जापान, हांगकांग, ब्राजील और कुछ यूरोपीय देशों में खांसी दमनकारी के रूप में विपणन किया जाता है।[1][2][3] इसे पहली बार 1972 में जापान में और फिर 1981 में इटली में पेश किया गया था।[4]

4-क्लोरोबेंज़हाइड्रोल [119-56-2] (1) का फास्फोरस ट्राइब्रोमाइड के साथ टेट्राक्लोरोमेथेन में हैलोजनीकरण करने पर 1-(ब्रोमोफेनिलमिथाइल)-4-क्लोरोबेंजीन [948-54-9] (2) प्राप्त होता है। एथिलीनक्लोरोहाइड्रिन (2-क्लोरोएथेनॉल) [107-07-3] (3) के साथ उपचार करने पर 1-(4-क्लोरोबेंज़हाइड्रिल)ऑक्सी-2-क्लोरोएथेन [5321-46-0] (4) प्राप्त होता है। पाइपरिडीन (5) के साथ प्रतिक्रिया क्लोपेरास्टाइन (6) का संश्लेषण पूरा करती है।
